I recently returned from a full semester abroad in Greece, and we traveled most of the country. Ultimately, it depends on how much traveling you want to do while you're there. If you've never been to Greece, the trip would be incomplete without a visit to Athens and the Acropolis, but some of the outlying areas are the most beautiful I've ever seen. Delphi is great, the monasteries (which I was completely uninterested in) at Meteora are phenomenal, Mani (mentioned in a previous answer) is different, but beautiful, and the island of Santorini, of the four I visited (Crete, Tinos, Mykonos, Santorini), was the most picturesque. Ultimately, Athens can be explored for as long as you'd like, but it can be extremely tourist-oriented, especially in some neighborhoods, so if you're looking for true Greece, you may need to get out of its capital.

Greece is a small country and if you have one whole month, you can see most of it, both mainland and islands!
Some places you should not miss on the mainlard are: Athens, Piraeus, Thessaloniki, Alexandroupoli, Metsovo, Meteora, Mystras, Dodoni, Crete's mainland.
And when it comes to islands, you should definately visit Mykonos, Santorini, Paros, Naxos, Kalymnos and if you can manage, visit also the Ionian islands in the West (Ionian Sea, between Greece and Italy).

Lucky you!
Have fun!

If you want a place that the tourists don't really know about because the Greeks keep quite about it then look at the second biggest Island ie Evia. About 2hrs from Athens by car bus or train.

Don't know about the best - I've only been once - but spent a wonderful 2 week holiday in Stoupa, on the Mani Peninsula (mainland Greece) - I can highly recommend it.

When exactly are you going there?

If you go before summer you should choose Athens or Thessaloniki. They are the two biggest cities and you will have a lot to see and do (sightseeing, tours, a lot of fun).

If you are travelling in summer try greek islands such as Santoriini or myconos.
