I hope you are not one of those perverts that plans to molest his "friend" while they are asleep. Whatever your reason, don't do it. Whether there is a safe way, there is no legal way to do it.

By definition, knocking someone out is harmful. You are either depriving their brain of something (air, blood) or rattling it around so vigorously that it shuts down for a moment. If it is a dangerous situation, like they are going to jump off a roof or someothing, and the result is more harmful than getting knocked out, then a choke hold is the best short of anestesia from a doctor. FYI, if anyone tells you of a knockout potion, don't believe it, that could really kill somone. A choke hold can also kill someone too, so it would have to be a very dangerous situation to use it. Basically though, you put your arm around the person's neck from behind, with your inside elbow bent around the front of their neck. Then using your other free hand grab the wrist of the arm over their neck and pull back enough to cut off their air. If you do it to hard you'll break their neck, and if you do it too long they could suffocate.

No, nor is there one to knock up a friend.
Seriously, if you mean physically, such as punching them or a strangle hold, then the answer is "no". The reason they would loose consciousness is because some physical trauma has been introduce to their bodies, even if was mild. Even the mildest of drugs can be harmful as any anesthesiologist can tell you.

Depends on how good a friend it is that you want knocked out.
I'm a fan of the round-house kick myself. If you want added oomph, try a spinning round-house. Or, if you prefer a more satisfying physical contact, you can try left jab, left jab, and right uppercut. Guaranteed to flatten the guy. And finally, to inflict maximum pain, you can resort to the side elbow strike to the temple!
Oops, you said non-harmful! Sorry, missed that part ;-)
