FAMILY GUY - The reason for this is simple - the creators of Family Guy took things from a number of TV Fathers - (Such as Homer) - That and the characters are great -
However, I am not taking credit away from the other two - They are great shows as well - But, I feel that Homer is the main character in Simpsons and the show revolves around him.
- South Park is great because they cover such touchy subjects and they don't care.
It all comes down to though, which one I laugh at more frequently. Which is Family Guy. That show is simple put - THE BEST-

South Park - they still seem to keep coming up with original ideas, even after all this time.
The Simpsons ran out of ideas 10 years ago, and Family Guy sometimes takes a "let's throw it on the wall and see if it sticks " approach, which can be pretty hit or miss.
Cartman still cracks me up after all these years....

In this modern TV era, adult cartoon comedies have fared very well with the 3 you mentioned as well as Futurama, King of the Hill and others. Simpsons has endured many years of popularity but I have barely watched it, but can see why people love it. South Park is full of funny characters but I have never watched it. Family Guy is one of the top 5 shows I have ever watched in my 48 years of life. It is a riot, it is well-written, has lots of flashbacks, movie and TV references, and has an overall twisted view of life. My humble opinion, of course.

definitely the simpsons because no matter how ,many times i watch the same episodes it always cracks me up and thre are so many obscure jokes that u dont pik up the first time so its always entertaining, ive been watching them since i 1st started watching tv im 22 now n i dont think il ever stop watching them,
btw.... you said which of the two but u gave us 3 to choose from?

They are all losing their sparkle. But I would say South Park because it has a little more edge than the other two.

The family guy. Peter is a lunatic. The baby stewie is crazy. The dog Brian is cool. Lois is a nut. Need I say more, and the crippled neighbor is hilarious. Sorry for got his name.

i love the simpsons. so real a guy--cares for his family, is lost, is trying hard to balance between being stupid and being a genius--my fav is bart. he is just wht truth is all about. he can put wise ideas in simple words, is a brat, yet he loves his sister and his parents. i wish i had him for a son.
