From Edward D. Wood Jr.'s 'Plan 9 From Outer Space'. It has to be the most pointless and stupid line in cinematic history. Just think about it for a second - the man was a moron.
The film is worth watching if you like bad cinema as is the Tim Burton biopic about the director 'Ed Wood'.

The film "Miller's Crossing" by the Coen brothers was full of funny lines, which is quite unusual for such a grim film. In the part where J. E. Freeman invaded Marcia Gay Harden's room, two armed men came rushing in to rescue her. Freeman shot them both, killing one and wounding the other. He asked the wounded man, "Leo sent you?" The wounded man answered, "Yeah, he told me to keep her out of trouble." Freeman said, "Well you did a real bang-up job! I'll be sure to tell him. By the way, where is Leo?" The wounded man asked, "How do I know that if I tell you, you won't kill me anyway?" Freeman answered, "Because if you told me, and I killed you, and it turned out you were lying I wouldn't get to kill you then!"

Most of 'Withnail and I' makes me laugh till I pass out, but particularly the bit where there's a rat in the kitchen, and Withnail is going after it with a manic, wild look in his eye, and he says:
'The f*cker will rue the day!'
Classic. Except it doesn't sound funny when you write it down. Weird.
