With the new TREK movie in the works, should new actors play these roles or should there be unknowns playing them?

I think unknowns maybe more believable.
Because this is a fun question I'm going to try and see who I'd pick to play them just for fun:
Kirk? How about Matt Damon!!! (strong, yet kind)
Spock? How about Keanu Reeves!! (he can have that serious intellectual look to him, and those "Spockish" eyes!!)

Kirk: Ethan Hawke.
Spock: Stephen Colbert

Nobody can replace Shatner and Nimoy. Nobody could be believable in those two roles. It was bad enough when Kirstie Alley didn't want to come back as Saavik (or however you spell it) and they got someone to replace her. It didn't work. Once you establish a certain person is a character, no one else can take their place unless you're a Trill and have a new host body.
