If it's plastic you can buy a blanking cap or screw on access cap.The main thing is don't pour wet cement down it as it could block another part of the system and don't block it too permanently in case you ever want to use it again

Take out the toilet and pipe to ground level then put in the biggest bricks you can fit. Pour in cement, smooth over and hey presto it's done

cut the pipe to or below ground level stuff the pipe with a newspaper plug so it is a tight fit to leave about 3" from top of pipe and then fill it with sand&cement mix

possibly through fact you're actually letting air into the equipment and that's why it quite is now working,i might look at my vent stack for a blockage and have the main line looked at additionally ok.
