I think that is such a tough question. For sure Life of Brian is up there, but is it funnier than Holy Grail? And for a non-python film Airplane has to be there. It's just totally gag-infested.
I also like someonelses choice, old school. I nearly died laughing(i'm not joking-it was scary) at Will Ferrell when he shot himself in the neck with the tranq gun. And of course Anchorman(check the avatar),especially the west-side story newscaster rumble, which is absolutley hilarious and incredibly daft.
I'm gonna plump for Airplane,with Anchorman a close second, with Life of Brian 3rd.And don't call me shirley.....

Dr. strange love.if you like Adam sandler or Jim Cary sorry no potty po po fart or belch jokes, this is pure intelligent comedy. peter sellers playing three rolls. George c Scott was great, keenan Wynn at his best. slim pickins just about stole the show in one of the most memorable scenes ever shot in a Hollywood movie.

while I agree that for OLD schrool comedy, LIFE OF BRIAN cn't be beat, I saw a fim called KUNG FU HUSTLE last year on a date,
the girl was into martial arts,I'm into comedy... we both we're laughing like alcoholics by 10 minutes in.
try it.. U'll like it !

What a way to open a film! Life of Brian is funny but was very Python formularised - The Jerk is stand alone hilarous. Good question.
