Electron microscopy looks at the structure of the specimen only and that structure is visualised by a highly artificial staining with electron-dense material. It would be foolish to make assumptions about function based entirely on structure and especially when that structure is being visualised by very invasive, artificial means. Physiology is about real, ongoing, complex systems - electron microscopy is only a snapshot.
