Chris Columbus said that he put seasons of luv at the beginning so that it would sort of take place throughout the whole film

In the play, Seasons of Love opens Act II and it's basically the last "happy" song people get before all the tragedy happens. Probably because Seasons of Love was the best known song from the broadway play in the film they wanted to lead off with that song because it was something that was easily recognizable. Filmgoers typically have shorter attention spans than theatregoers but also in a film you sometimes have to tell the story differently than you would in a play and vice versa. All in all, the reason Seasons of Love was put in the middle of the play and at the beginning of the movie was most likely a choice that the creative team felt helped best serve the story.

The song was placed at the beginning of the second act because typically you need something catchy to bring the audience into the action after intermission. It was not at the beginning of the show because it doesn't really have anything to do with moving the story along. SO they went directly into the plot at the start. But having it at the beginning of the second act allows those people straggling in late not to miss anything.

What are you talking about, without the film? The movie and the play are not the same thing.

The song occurs a few times throughout the play, not just in the middle.
