Where do I start, will be a big show so I want to make it the best possible.

This is just a monumental question. You say it is going to be a big show - define "big." Are you talking about 50 quilts, or 500? Is it a local guild show, or regional? How many attendees do you expect to have?

This will not answer your question, but at least it will give you something to start thinking about.

1. Create a timeline and stick to it. It take a lot longer to organize something like this than you probably think. You will also need to create a budget.
2. If you are going to have vendors as well as quilts on display, start sending out feelers as to which vendors you want, and if they will be available on that date (or dates). Are you going to charge a fee to the vendors, and if so, how much?
3. Determine the categories you are going to have for quilts and what the requirements are. Decide on a deadline for applications. Create forms and make them available to your quilters.
4. Will you be providing food? How? Who is going to make it - a caterer, your church group, the guild, the facility where the show will be held, the local scouting group, etc.
5. Where are you going to hold the event? Schools are great, as are community buildings, but if it is larger, you will need a larger venue, and that could cost.
6. Do you have a way to display the quilts (frames, etc.)? Do you have volunteers to help with setup and tear down, and white glove ladies to handle the quilts?
7. What will you charge for attendance? Are you doing one or more prize drawings?
8. Who will judge the quilts? You want someone who knows what they are doing as a JUDGE, not just as a quilter.
9. What are the awards for the winning quilters? Will you have a viewers choice award? How will you organize that (slips of paper with numbers on the quilts)?

If at all possible, find someone local who has done this sort of thing before and pick their brain. Check with your local quilt guild, or a quilting shop. There are 87,000 other things you will need to think about, but this at least might get you started.
