Understand the profile they have asked your resume for first. Include the refrence in the subject of the email. In the body just tell them about where you are working currently and that your resume is attached.
Attach the resume. In the resume have a very strong paragraph on your career summary which should give a clear idea about your current profile.

A cover letter let's you express your interest as well your strengths and prior experience if you have any. It's nice to include with your resume. Good luck.

Send the resume as an attachment. Your reply should be a cover letter of 20 lines or less, with one line that shows you know something about their company, then your promise to follow up on such-such date. then make sure you do it.
