If your boss hasn't left you specific instructions on what to say you can either say:

He/she is in a meeting right now. Can I leave a message? (Or "Is there something I can help you with?" If that is appropriate and there is something you can actually help callers with.

or

He/she is unavailable now, but will be back in (one hour, half hour, etc.).

Always offer to take a message and don't make any promises that your or your boss cannot keep!

Unless your boss left specific instructions on what to say to any callers, you simply let them know that your boss is in a meeting, and ask if you can take a message.
