Studies show that when you say something to somebody for example "you're fat" or anything of that nature, and you say you mean it as a joke, 75% of the time it is actually isn't a joke. So yes, 75% of the time it is true.

No "ALWAYS" is an absolute. In order for your question to be true, no joke anywhere, anytime, written or told by anyone, in the history of human humor could have been executed without truth.
Truth: Two guys going bear hunting, one puts on his boots while the other his running shoes. Boot guy say, what do plan to do, outrun a bear? Running shoe guy, no, I only have to out run you.
No Truth: How do you catch a polar bear?
Cut a hole in the ice, line the edge with the contents of a can of peas. Hide and wait. When the bear comes down to take a pea, kick him in the ice hole.
I was in Alaska for three years, the first one has an element of truth. The second on the other hand is just silly with a bit of play on words.
I will grant you a great deal of humor has an element or thread of truth woven through it. But "ALWAYS" is just too strong a word.

ALWAYS is an absolute, so I doubt it. However, there is definitely some sort of familiarity in a joke that makes us laugh. A joke very often is a play on words, which requires some recognition of double-meaning in language.
