From what I understand he was shot in the back while playing poker..
the legend of aces and eights being the dead mans hand comes from that's what Buffalo Bill was holding when he got shot.

edit:

The inaccuracies on this page take your breath away.
Jesse James was shot in the back of the head while straightening a picture on the wall. It was Wild Bill Hickok (not Hancock) who was shot in the back (of the head again, I think) while playing poker. (He had a hand of aces and eithts, which has become known as the "dead man's hand"). "Buffalo Bill", Wm Cody, died in his bed well into the 20th century. He was actually quite a bogus character, really a very minor figure in the history of the West as opplosed to its selling. My grandfather met him once, but so I think did quite a lot of other people's.

Chefzilla is thinking of Wild Bill Hancock. William "Buffalo Bill" Cody essentially died of old age.

Moderatep is right. It was Wild Bill Hickcock that was shot while playing poker. The "Dead Man's hand" originated with him. Aces and eights.

That would be, "How did Buffalo Bill DIE", not died.
