Definately the Quiet Man and Red River and She wore a Yellow ribbon...heck all his movies were great! There's one on BBC1 right now!

Stagecoach (1939), 96 minutes, D: John Ford One of the great American films, and a landmark in the maturing of the Western, balancing character study (as disparate passengers travel together on the same stagecoach) and peerless action (in a lengthy Indian attack, featuring Yakima Canutt's famous stuntwork).
Also the film that propelled John Wayne to "genuine stardom". Mitchell won an Oscar as the drunken doctor, as did the music score. Script by Dudley Nichols, from Ernest Haycox's story "Stage to Lordsburg" (whose plot is reminiscent of Guy de Maupassant's Boule de Suif). Filmed in Ford's beloved Monument Valley on the Arizona-Utah border.

Jeez, he made about 70 movies, that is not easy.
I would list Stagecoach, Red River, The Man Who Shot Liberty Valance, True Grit, The Shootist, The Quiet Man, The Searchers... and that is just for openers.
A very underrated actor. All those films I listed are pretty damned good too.

With John Wayne, it's not one movie. It's the character he played. "The Duke." He played that character in most of his movies, and it's that which he's famous for.
I like the Quiet Man best, because he's not the Duke - he's a new character.

The is no one movie..........
His career spanded over 50 years of films. His John Ford films are some of his most noteable. My personal favorite actually is not a western. "They were Expendable," about PT boat in the Phillipines.

The Alamo, because he dies like he does in all his films. I met him once filming around Tower Bridge, he was a really nice guy, but wooden at acting.
