Multiplex is a term for TV cable systems that broadcast with different time zones, for example HBO west and HBO east with 2 hours difference, this is called MULTIPLEX channel.

Most multiplexes are modern theatres with Dolby Digital/THX/Surround systems. More modern ones have digital DLP projectors which really bring out the colors very well and the brightness and dark levels are superb. This is hard to find in a single theatre. Most modern cineplexes have stadium seating, and the really good ones have a really steep stadium arrangement, which is a must. Having someone's head in front of you is the pits. However, a major distraction in movies, whether in a cineplex or an isolated theatre, is the awful habit of people, especially Indians, adding their own soundtrack to the movie. I just can't stand this- it takes away the pleasure of watching a movie.

No difference, unless you can sit in more than two cinemas at the same time! Of course, multiplexes tend to be newer.

It is. Just a matter of preference, really- stadium seating is usually a big draw for movie plexes.
