here is the safest way. tip: your systems boot up files might have spyware or viruses so don't scan your computer after you load windows.
1. if you have a virus scanner and a spyware remover update their files from the web.
2. turn off your computer. go around the back and remove whatever cable is used to connect you to the internet.
3. turn your computer back on and hit F8 a couple of times before windows opens to get into the safe mode.
4. now scan for viruses and spyware.
5. turn off your computer and back on again. now all of your spyware and viruses should be gone.

Try Trend Micro Internet Security 2007, its anti virus anti spyware anti malware anti adware firewall and everything all in one program. You can download a 30 day trail from the link below.
I will be buying it for sure when the trail runs out.

If you have tried both AdAware and Spybot and the thing is still there, the only real solution is to reformat your computer and do a clean install. However, before you do this, you must save any files or documents that you want to keep on a CD or USB memory stick.

Go to Tweak XP and follow everything they tell you to do for your operating system. Make sure you have time to do it as it will take awhile.
What they have you do is download freeware and shareware that will clean and protect your computer BUT you must take it step-by-step as they tell you.
I had a particularly nasty virus that nothing else would get rid of and after I did as they said, I haven't had a problem since so I heartily recommend them.

download different anti-virus scanners and ant-spyware (Spybot Search & destroy is a good one).
If worst comes to worst, save all the files you want to keep on an external harddrive or transfer them tro another computer, then reinstall your operating system.
