I found a few movie plots that claim to have the homosexual characters, or lifestyle!
It all depends on weather you are looking for main stream or independent films.

I like Samiracat's answer the best.
But, I did want to add that there is was a special about Broadway, and in there, they tell that La Cage aux folles ("Birds of a Feather') was the first homosexual broadway show to make money! I believe that was around 1985 when I saw it. The Birdcage was based on this show (same writers/producers), but I believe the show is based on a much older French movie called La Cage aux folles from 1978.

You might be interested in Richard Barrios' book _Screened Out: Playing Gay in Hollywood From Edison to Stonewall_ (Routledge Press: NY, 2003).
The author points out that a lot of early film was lost, but it LOOKS like a 1912 film called "Algie, the Miner" might be the first one to portray a "gay" seeming character---he's got a girlfriend, but apparently is camping it up like crazy as a really effeminate character....
The book is a good read, with fairly decent references.

omg i have no idea... but the first movie to come to my mind reading this question was ALEXANDER (with Collin Farol in it) that was the gayest movie i have ever seen! (litterally speaking it was)
