You should probably let your boss know that you'd like a few minutes to talk to him or her about something privately. When you are alone, you can mention that you've discovered that there's an opening within the company that you'd like to pursue.
Hopefully your boss is already aware of the opening, and if not, you might want to explain what it is and how you feel it would best fit you. Hopefully your boss will be supportive and may even give you a recommendation for the job. Companies usually go for internal candidates for ease of training, so you might have a good shot!
Try to do it as soon as possible so your boss will have time to find someone to hire or divide your current projects accordingly. It's only courteous.

Be upfront about it. Tell him that even though you may enjoy your current job, you feel you need a new challenge and a chance to expand your knowledge and skills. Most bosses can get on board with that.

As a boss my self I will always prefer a direct approach, but it is a good idea that you know all that there is to know about that job that you want.
