I am going to Japan on saturday with work. I have 2 days free in Tokyo, 2 days in Osaka, and 2 days in Kyoto. What things in these 3 cities must I definately NOT miss !!?

In Tokyo, I recommend the Imperial Palace, the Tokyo Tower, and Meiji Park. Also walk around Ginza or another huge shopping area.

In Kyoto, I recommend Nijo Castle, Kinakuji Temple and I also think you should just walk around. It is an incredible city!

I've only been to Osaka once and don't remember anything special so I can't give you advice.

In Kyoto, must visit Kiyomizu temple and Kinkakuji temple.
In Osaka, must eat Okonomiyaki and Takoyaki.
In Tokyo...go to Kaminarimon to experience good old Tokyo and eat at a little sushi bar and a tempura bar. And then go to Shibuya for watch people and shopping.

In Tokyo, I recommend to take one of those "Hato Bus Tour". I took my husband as well as many of English speaking friends, and they all love it. Don't forget eating authentic sushi in Tokyo. It is the mecca of real sushi, not cal-roll or spicy tuna roll. (be aware that good sushi is not cheap. It will cost at least 10000yen P.P. ) At Tukiji Market in the early morning, say 5am is a great spot to eat one of the top notch sushi only at 4000yen. It worth to wake up early to line up.

To go to Osaka from Kyoto is only 1 hour train ride. People actually commute every morning. Osaka has 2 down towns. One is in north side of Osaka called Umeda, and the other one is in south called Shinsai-bashi. Only 10min subway ride. You may see some very funky looking kids there.

In Kyoto, It is good idea to join a tour because they cover major tourist sites. Kyoto has a number of UN WORLD HERITAGE sites, wonderful Zen garden etc. If you have a chance, go to Gion area at night time. You may run into authentic Gisha girls walking down the street.

Basement of every single major department stores is a lot of fun for quick bites. Warm noodle, bread, sweet, bento box....etc.
