Actually, employers can ask any employee to take drug tests wether they suspect that the employee is under the influence or not. Many employers do random drug checks on their employees as a matter of course.

Random drug testing of employee by employer varies from state to state. I can tell you that if you are a public servant or involved in public transportation the answer is likely "yes". However, there is a legal arguement to follow if you want to upset and change the laws of your state. It's your body and you will not give part of it away without being compelled by a court. Of course, by the time that happens, you're clean. The employer can't fire you or discriminate against you for exercising your rights - unless they want to face a civil suit from you for millions. Anyway, in most cases, the answer is going to be "no" simply out of practicality. However, if your work contract (if you have one) states that you will submit to one if asked, they may have the right to let you go if you refuse.

They should be able to. The employee is on their payroll, their premises, probably, and represents their employer.

I would say its up to company policy. Then again, they could always just fire you for being intoxicated at work.
