you need Photoshop to do this and for it to look professional. You scan in your drawing as a bitmap file then convert it to cmyk in photoshop. then you make your line drawing the top layer (first selecting the white and deleting it) and then you can fill or colour in the layers beneath it. your bottom layer must be whatever background colour you have chosen. for more info go to photoshop help online,

Try a Wacom tablet. It allows you to draw as you would with a pencil with the ease of a computer. Once a drawing is scanned in it can be used with the tablet. I use mine in conjunction with Adobe Photoshop for coloring and adjusting.

Download this free software called art rage. It's great! You can colour in any picture with some practise. The free version is quite good, but not all of the tools are available:

i believe there's a special software program you can buy to do that. i remember we had something like it in my art class in high school. Im not sure what it was called but if you go to an Aaron Brothers or any art store im sure they could tell you if you explain what your looking for.
