A BIOS boot password on laptops are the most effective, otherwise just a windows password.

I would like to add to reguser2005's answer the following:
What he suggested (which are all valid) can still be bypassed, so even if there are no cheap ways to get better security. at least you can take some additional measures to find out if someone tampered with the system. So here are a few more things to do:
1. Lock the case of the PC. If not the BIOS password can be removed.
2. Use case intrusion logging in BIOS to find out when was the last time the case was opened (if supported).
3. Enable login auditing in Windows (or whatever OS you're using).
I would have some more suggestions but those are becoming probably too complex for the regular user.

I am assuming that you want to restrict login access. You can password protect your machine at multiple levels.
a) enable boot time password. This will restrict users from accessing your BIOS as well as OS
b) Enable Hard disk password. Many laptops and desktops now allow hard disk password. If your computer is stolen, or accessed, you can be sure that the data cannot be accessed unless they have hard disk password
c) Enable Operating system password. This will ensure that your documents and files are not accessed without proper authentication

A couple of methods:
1. Enable BOOT password in BIOS
2. Use a OS like Windows NT/2000/XP/2003 that supports stronger login mechanism
3. Use a complicated login / password that no-one can guess.
4. There are hardware based login devices available (finger-print scanner) that ensure that only you can login.

If you have win XP you can add a password to your user.
Just click in the small pic on top of your start menu and press back at the new window, there you'll find the option to add a password, enter it twice and press OK.
Now every time you'll open your PC it'll ask you for your password in order to continue.

Set up a password so when you turn on the computer it asks for a password. Make sure its a really hard one with letters and numbers that no one can guess

Do you mean opening or using?
A password will stop people using it, to stop them opening it, fit a lock. Most cases have a bracket where you can fit a padlock.
