I think my favorite comedy movie in the world would have to be "Little Miss Sunshine." It's an... interesting movie. The girl wants to go to the Little Miss Sunshine Pageant (which is something like... Miss America Pageant for little girls.) *even though she is chubby and... well... not... pretty* So her entire family drives to California in their crappy car. Each of the family members happen to have a weird flaw. The father is crazy about winning. The mother smokes when in stress. The grandfather is the little girl's coach and does heroine. The uncle is suicidal, and worst of all, her brother took an oath of silence so that he won't talk until he joins the Airforce. Funny movie with smuggling dead bodies across statelines and pornography saving them from getting a ticket. A must-see movie!

Too difficult to choose just 1. I think different time periods have their movies. I absolutley love Singing In The Rain, it's kind of a comedy musical. Then in the 70's, I remember standing in a long line to see Stir Crazy starring Gene Wilder and Richard Pryor. I thought I would die laughing in the theater that night. Then when the Nutty Professor, with Eddie Murphy came out, I was again doubled over laughing. I was hysterical while watching White Chicks too. I think I would have to say, over time those are my favorites.

"What's Up, Doc?" with Ryan O'Neal, Barbra Streisand, Madeline Kahn (rest in peace, she died way too young), and a host of other funny people. The whole film is hilarious, but the chase scene filmed all over beautiful sections of San Francisco near the end is also one of the best staged scenes anywhere, in any film.

at first i was going to say its a 3way tie between Blazing Saddles, Wedding Crashers and Eddie Murphy's Delirious. but hands down its, "Uptown Saturday Night" with Sidney Poitier, Bill Cosby and Harry Belafonte...hands down.
