Here is how I understand it:
You have to gather a lot of energy (get up) to have a great time, impress people, or be the life of the party (get down).
It could also be construed as the poster immediately above me stated: a male would have to get an erection ("get it up"), in order to have sex ("get down").
In the drug subculture, it could also be understood to mean that you must get high (use drugs; get up) to truly understand or accept things ("be down with it").
Depends on context.

Means if you want to be comfortable say in life,you have to get up and work then after you can get down and relax.
