Not knowing the kind of business makes it more difficult to give specific answer. That being said I would advise you to treat the interview with extreme professionalism on any question regarding the business. In other words, have a no BS line of thinking when it comes to the business and return of investment.
If the interview leans on anything private or not having to do with business, go light, well placed humor works if: everyone understands the wit.

Stay relaxed and be prepared. Research as much about the company as possible and let this be manifested in your conversation. For really helpful advice that has always led me to success check out about.com. Just type in interviews. It will query up a wealth of success-proven tips and information about interviewing.

You have to soft sell yourself and be informed about the company. He is only a man, don't let his position intimidate you. Tell him why you want and deserve the promotion and how you enjoy working there. Be well groomed, of course. Don't forget appropriateclean shoes, manicured nails and clean, styled hair. Not too much makeup and no perfume.

One thing not mentioned is to dress for the part. Do the people dress more business like or more casual than you do in your current role? Dress like they do if you don't already.
Also we like to hire people that will fit in. So if the group is laid back a little try to be yourself while keeping it professional. If the group is more stern and serious be that way yourself.
Good luck!

Practice with a friend or family member. Sit down with them and do a mock interview. That will give you some confidence before you actually do the real thing.
