Iodine in the Lugol's when it heavily stains plant material is actually fixing (killing) the tissue by adhering to the starches, etc. You may still see some cyclosis in the lightly stained plant leaf cells.

Lugols is mostly iodine with potassium iodide. Pretty useful after a nuclear accident.
