The TKTS booths are great for the day of the show, especially because you can get the tickets half price. The main booth is currently located at the foot of the Marriott Marquis (47th and Broadway) while the buiding is being redone. Many people don't know about the second booth, downtown, Lower Manhattan Theatre Center at South Street Seaport (the corner of Front and John Streets, the rear of the Resnick/Prudential Building at 199 Water Street ) which sells the tickets the day BEFORE the show (so you can buy Wednesday matinee tickets on Tuesday). It's easily accessible by subway (plus a short walk). See the tdf site.
Right now, Off Broadway, Altar Boyz has gotten very good reviews (it's a musical), as has The Hairy Ape (straight play), and Slava's Snowshow (not sure how to describe this one, a little out there, and ends with the audience in a "snowstorm"). The last play is closing soon. The Fantastics played Off-Broadway for 1000 years, and is now back, although this new company has recieved only mixed reviews. Still, it is a charming show, and many consider it a hallmark of Off-Broadway.
All that said, sometimes it is fun just to see something you have never heard of (especially if you can get the tickets half-priced), as you may get to see something amazing.

If you aren't picky about what you see, the TKTS booth is good and has discounted tickets ( http://www.tdf.org/tkts/ ) . There are usually some Off-Broadway shows up there, but it varies from day to day. You could also try just going to the theatre and seeing if any tickets are still available.
