Although most answers are good, they either are only referring to a specific case, like in the world cup, the New Zealand Japan game is the highest, whereas others have said the Argentina Paraguay was the highest. While I do not dispute the Argentina Paraguay as the highest, I found this site that list matches that have the winner has scored more than 100 points.

Argentina 152 Paraguay 0 in May 2002 remains the biggest winning margin ... beating the previous record by a single point.
That was in 1994 when Hong Kong beat Singapore in a test 164 - 13 ... winger Ashley Billington scored ten tries in that game. That remains the biggest scoring game.
Third biggest is England 143 Romania 0, November 17 2002, Twickenham when Charlie Hodgson scored 44 points.

new zealand has highest rugby union scoreline in world cup cant remeber who playing think it was japan .i know marc ellis scored 9 tries in it .score was 154 to 17
australia got 2nd highest score versus namibia 140 to 0
