Roger Federer already sure can hold his No. 1 position. However, for the ladies, i think it is the fight between Amelie Mouresmo and Justine Henin-Hardene.

Roger Federer has a lock on the World#1 ranking on the men's
side. It doesn't matter what Nadal does between now and the
rest of the season, Federer will end the season at #1. He clinched the top spot for the season when he won his third consecutive US Open last month. Amelie Mauresmo has the lead right now, but Justine Henin-Hardenne and Maria Sharapova are within reasonable striking distance of the lead. Either one of them could overtake Mauresmo and catapult to the number one ranking by the end of the season. I think the rest of the field can forget it. If Mauresmo doesn't end the season at #1, it will because JHH or Maria will be number one. I think the other players are too far back to catch Mauresmo.

Federer is pretty much guaranteed to end the year ranked #1, so the question is more whether or not he'll win the Masters Cup. Having won it two of the past three years, he's certainly the heavy favorite, but it could be tough. As dominant as he's been, his form on indoor courts has been a little patchy. He stormed through the tournament in dominant form last year, only to blow a two-sets-to-love lead against Nalbandian in the final. If the Argentine is there again this year, he could pose a threat. He is one of few players who still has a winning record vs Federer. Another tough possible opponent is Nadal. He's looked a little shaky of late, but a 6-2 head to head record against the world #1 puts him in as a strong candidate. Ljubicic will also be worth watching. With his huge serve and precise power game, he can be a handful for anyone on a good day. James Blake could do some damage too. His fast paced aggressive game could trouble anyone and, this being his first appearance at the Masters, he'll be keen to make a good impression.
On the womens side, it's a little more interesting. Unlike the men's game, the top spot is very much a three-person race. Mauresmo, Henin-Hardenne, and Sharapova will all be going in on the back of banner years, and will all be eager to cap it off with a win and a #1 finish. If Clijsters shows up injury-free, she'll also be looking to make some noise. Having surprised many with a return to the top-ten, Hingis will be keen to show she's not there just to make up the numbers. Should be a very interesting couple of tournaments.

Roger is alredy guaranteed to end the year at number 1. In the womens I think Sharapova has the biggest chance to dethrone Mauresmo mainly because she has no points to defend at all. If she wins Zurich and Linz she could probably overtake Mauresmo, she will earn 600 pts. if she does so. Mauresmo has a title to defend and would probably be sidelined with her shoulder injury. I also think that Henin has no chance of ending the year at no.1 because she hasn't played since withdrawing in the Fed Cup final and will go directly to Madrid not being 100%.

I would say Roger Federer and Justine Henin-Hardenne will end the year as World No 1. Not Amelie Mouresmo and Rafael Nadal.

I think it will be a close match between Mauresmo and Henin Hardenne
Personaly, I'm cheering for Sharapova. I think she has a good ahot at No. 1 if she keeps up that big game she's been playing with.

No I think Roger definitely can, but Mauresmo has too many good players just below her (Sharapova, Henin-Hardene) that are probably going to overtake her by the end of the year

Roger is good to go for the rest of the year, I don't think Nadal can win enough points to beat Roger this year, he is way ahead, but I don't think Mauresmo can keep her spot, shapova looked pretty confident in the US Open!
