Both the roads that you have mentioned are wrong because it will take you longer time and more distance to reach mahableshwar by both these roads. If you go to mahableshwar via bombay puna road then you will have to travel at least 305 kilometres between mumbai and mahableshwar. And if you come to mahableshwar via nasik puna road then again you will have to cover even more distance because the distance between ahmedabad and nasik is much greater than the distance between ahmedabad and mumbai and also there is no direct road between ahmedabad and nasik.
The fastest road to mahableshwar from ahmedabad is the ahmeadabad-mumbai-mahableshwar road via mahad. This is the shortest, fastest and direct road from mumbai to mahableshwar which is only 237 kms in total distance. In case you are not aware, mahad is a town that lies along the mumbai-goa highway which is also called as National Highway No.17. So all you have to do is take the mumbai-goa highway which starts from panvel at mumbai and you will be on your way straight to mahableshwar via mahad.

Best possible travel plan for u will be:

Please travel to Mumbai by railway. It's fast and easy on the body.

From Mumbai u can take a volvo bus and stop bothering about which route to take.

Or u can book a vehicle and take a route via Mahad on Goa highway. From Mumbai the route will be as: Mumbai - Panvel - take Goa highway - Pen - Wadkhal Naka - Nagothane - Kolad - Indapur - Mangaon - Mahad - Poladpur - from Poladpur leave the Goa highway and take a state highway to Mahabaleshwar- U can stop to visit Pratapgad enroute , a historical fort of Shivaji - keep continuing in Ambenalli ghat section and reach Mahabaleshwar.

Now a days the roads have been widened a bit.
