Yes, you can have 2 drives on your computer. I assume you have a CD drive as well? D: is either another hard drive or your CD drive. Look and see what is on the drives and do a manual cear out of data only - just get rid of anything - delete it - that you don't need. then look at programs in remove programs in control panel. Your computer needs a little room to do the defrag. If you have a Windows disk - it depends which version you have - each vrsion tends to reformat differently. If you don't get it working - ask the question again and tell us what the operating system is 98, ME, XP what? Do you have a CD drive or CD writer/ re-writer? It may help to know the make or even if it's a desktop or laptop.

Formatting a drive should be done only as a last resort only, because it erases everything on the drive and requires reinstalling everything on the drive again.
A CD Drive would be identified as drive D: If you don't have a CD drive, it is probably a partition on your drive C:
You can gain a lot of space by doing disc clean manually. Start, Settings, Control panel, Internet options, Delete Cookies, Delete files, Erase history. Next to History, set it to keep History for only 1 day.
Go to C:\Windows\Temp and delete all the files. There may be 2 that won't delete, don't worry about them.
Go to C:\Documents and settings. click Tools, Folder options, View, and click show Hidden files. All the users should be listed in their own directory. Open Owner, or the owner's name folder.Open the Local settings folder and delete the contents of these folders: Temp. Temporary Internet files(if any are left) and History. Do not delete anything in the Application data folder.
That should gain you a lot of space.
If it just losing space without you adding to it, it may be infected with spyware, adware or a computer worm. Get a good antivirus program such as Norton and clean your drive. A free antivirus can be downloaded here:

Delete some files you do not need...also, you might want to try using CCleaner, it will do a disk cleanup and more.
