Well it depends on what type of email you want to do this with. If you are using Outlook or Thunderbird it's pretty simple.
1) Go to Start > Control Panel > Sounds and Audio Devices
2) Click on the Sounds Tab
3) Click on New Mail Notification Tab.
4) Browse for the sound you want to use for your mail program
5) Hit Apply
Viola...You're all set.

In outlook / outlook express, go to tools - options, and set the audio settings on. Better still, download incredimail - it has a notifier with audio / video.
