If you mean setting her up with her own user name that's viewable when your computer starts, then yes. Go to "User Accounts" in control panel, click on "Create a new account".
If you mean adding her to an existing username (e.g. if your user name is "husband" and you want it to be "husband & wife") Go to "User Accounts" in control panel, click on your username, then click "Change my name"

if you mean as a user, yes you can, but if you mean to have both of you signed in at the same time on the same computer on one account name, no, that is not possible...
