Likely best to leave out of NY to Puebla, Mexico.

Ok, if you drive non-stop, it will take you about 40 hours and 50 minutes. You need to pass through the border in Laredo, TX. In the Mexican side you will get to Nuevo Laredo, Tamaulipas, and then go all the way down to Puebla City.
PS. For driving directions in US I suggest www.mapquest.com, for driving directions in Mexico I suggest www.sct.gob.mx, then click on the "Traza tu ruta" logo. It's in Spanish only though (obviously), but for the record: Nuevo Laredo is in the Mexican State of Tamaulipas and Puebla City's state has the same name (Puebla)
