The point or main idea that the reader should be left with.
Generally the theme is a message that the author is trying to convey to the reader. It can be presented as an underlying truth behind a story (seen in fiction and poetry) or blunt and to the point (seen in textbooks or news.)

The theme is the main idea, message, or point that the author is trying to make. The author might make that point with smaller themes or ideas that run througout the book. For example, a book might have the theme of "redemption" but also the related theme of "sin" or "error." A book might have the theme of growing to maturity, but have a related theme of hindrances to that maturity.
If you have to write about the theme of a book, look for something that is demonstrated througout the book. Write down specific examples that demonstrate that this is the theme. The major theme should be something you see at the beginning and end of the book as well as in the middle. You will usually find it in the introduction and then repeated or reflected at the end of the book.

The message or moral
Example: In To Kill a Mockingbird, treating people fairly regardless of how different they are from you, is the theme.
OR
A common thing throughout the books
Example: In Harry Potter, weird little things like socks are what are called "running themes" (they're mentioned throughout the books, and carry with them some meaning)

The theme is the main idea. If you had to tell someone, in one very simple sentence (without the cool details) what the whole thing was about.
