I really like Tristan's answer, above, and voted his response as best answer.

However, for my money, it is Arthur Miller. No one that I know of has been more masterful in discussing universal issues, and universal truths. "Death of a Salesman" is about us all - and "The Crucible" is about what our government is trying to do to us today.

It really depends on who you ask. Even considering Shakespeare to be number one is questionable in my opinion. Different people say different playwrights. For example on one list (*) Dante Alighieri is number two. However, I don't know many people who'd agree.

In my experience, it usually comes down to Ibsen or Chekhov. I think they're both superb, but there is something magical about Chekhov that would elevate him to #2 on my list. Ibsen's plays are remarkable for their realism, and their author's bravery in examining issues that were absolutely scandalous when the pieces were composed. But Chekhov's major works, when done well, have an air of timelessness about them. They seem to be not only about the characters in the play, but about ALL people in ALL times.
