Its about an 1hr 20 min flight about 550 kilometers,
I was think of renting a car in Istanbul and leaving it in Athens (if that is do able)? Should be able to drive that in 8 hrs? I'm planning this for next May.

its about 1.200 kilometers not 550,you will definitely need more than 8 hours

You can ask a car rental company (I would suggest Sixt Rent A Car).
Although Greece has a very good road network, it depends on the way you drive.
I have driven from Alexandroupoli in Greece, close to the Turkish border to Thessaloniki in 3 hours and from Thessaloniki to Athens in 5 hours, but I was speeding a lot, sometimes exceeding the speed limit.
You should estimate a 9 hours drive without stop from Alexandroupoli to Athens and you should add the drive from Istanbul to Alexandroupoli (I have been told that it is approximately 3 hours).

Well,550km is from Thessaloniki to Athens,so it's more.A 2day trip is what I recommend(with several stops).It's a beautiful season(no rain)and you can visit really nice places.You can rent a car for sure.
