in the absence of any visible abrasions or sores or other signs of injury, I would venture a guess that you are circumcised and quite possibly were cut a little tighter on one side than the other. While the shaft skin will stretch to accomodate such an occurence, you may be in an early enough stage of puberty to not have had enough time for it to stretch out.

maybe a thin spot in a vein, you might need to see a doctor about that, does your penis curve when its erect?
if so thats a different problem.

you should prob get tested for stds & also go to your doctor for a routine checkup.. who knows.. maybe you're just having growing pains =)

it could be a blood clot, either way talk to your doctor about it next time you see him. im sure its nothing serious.
