It can be rude to assume that someone's interested in you and drop the "I've got a significant other, so back off, boyo" thing. You may be misinterpreting someone's interest or friendliness for actual romantic interest (believe me, everytime a girl working on the counter at McDonalds makes eye contact, I wonder if she's just hitting on me. I think straight men have ruined the service industry forever.)
I've had conversations with people who have slipped in the fact that they've got a significant other and I've been greatly insulted. It's like "I know you haven't made a move, but I want to nip this in the bud before it even gets started because I find you repugnant."
There are gentler, kinder ways to slip the words "my boyfriend" into a conversation without it being blatant. The secret is to wait for the exact moment in a conversation that you can slip the words "my boyfriend" into a sentence and still make it sound natural.
What to do:
"So, what did you do last night?"
"My boyfriend and I went to the club, the new one on 54th street. Have you been there yet?"
What not to do:
"So, what's your favorite type of food?"
"Well, my boyfriend and I like italian food. Well, I think I like it because my boyfriend's italian. My boyfriend thinks I cook well, but I think my boyfriends wrong. My boyfriend. My boyfriend. My boyfriend."

The gender of your interest shouldn't be a factor when formulating an answer.
My boyfriend was actively pursued by a co-worker who wouldn't leave him alone until he said, "I am currently in a committed relationship. I am always happy to have more friends, but if you're looking for more than that I am not sure my partner would understand." It worked for him - good luck!

You could just say that you are involved in a relationship right now. You could just avoid the issue altogether and change the subject or you could just say I am too busy.
