The road distance from Mumbai to Trivandrum (Thiruvananthapuram) will be 1,543 kms. The route I would suggest you to take is as follows:
From Mumbai, go to Vashi and then Panvel. From Panvel go on the Mumbai-Goa highway which is also called as National Highway No.17 It starts from Panvel and goes all the way to Edappalli which is near Kochi in Kerala. This highway is a straight and direct route to kerala. It passes through goa state and Karnataka state along the coastline and finally ends at Edappalli. From Edapalli to Kochi there is a short and straight road which should not take you more than 2 hours. There are plenty of directions along the road to show you how to reach there. Once you reach Kochi, from there is an entry into National Highway No. 47 which will take you straight to Trivandrum (Thiruvananthapuram). There are enough of signboards along the way to show you how to go there so don't worry.

If you are travelling in train please catch Mumbai - Nagercoil express it will reach nagercoil with in 38 hours, from there Trivandrum will be one and half hour. distance may be 2000 KM.
This is easiest.
If road means go by bus Mumbai to Bangalore and then from Bangalore to coimbatore then from there to trivandrum.
