There are several ways. Radar is a big one for tracking, but reports that come in from Skywarn Spotters, Law Enforcement and then general public is the most useful for collecting information on damage.

The use a Doppler radar to detect rotation in the storms. They can watch the "rotation" (which does not necessarily mean a tornado has touched down but is possible) and track the direction and speed of the storm. The actually tracking of a "physical" tornado is relied upon spotters.

Radar mainly. They often go in the vicinity of the tornado in a van containing many different instruments that are usually connected to a computer to collect data. They're called "storm chasers".
