It is about a plane crash on an island that seems to have been used in the past as the venue for some sort of scientific experiment by a group called "The Dharma Initiative".

I won't give away too much, but there are supposedly these people called the "Others" who cause trouble for the survivors. If you haven't seen season one yet, you will be confused about the second season.
