I assume you are doing chromatography.

For paper chromatography the rf value is the ratio the spot moved compared to the distance the solvent moved from the starting point.

Rf=distance spot moved/distance solvent moved

Simply use this simple formula
Rf Vin = Vout R1

Rf = Vout R1 / Vin

With that you get Rf

Measure the distance travelled up the plate by the spot of the substance, and divide it by the total distance travelled by the solvent from the moment it got to the original location of the spot.
