The two best equipped shops that I have found (and my favorite needlework places in central London) are Liberty 3rd floor. What and Aladdin's cave that place is for me.
And
John Lewis the ground floor in the haberdashery department. There are wonderfully helpful ladies there that assist with wool choices and pattern deciphering!

Try the bid department stores - I know that John Lewis sells knitting wool (individual balls and large packs of about 12)

The following shops all sell knitting yarn:
Liberty (on Regent Street),
John lewis (on Oxford Street),
Peter Jones (Sloane Square)

This web site provides useful reviews of all of them:

John Lewis is very good. Liberty also but very expensive.
Worth trying out internet for Texere Yarns. You can buy a catalogue from them for a few pounds and it includes samples of all the wools - amazing selection.
