Though there are plenty of all-in-one solutions (firewall, anti-virus, anti-spy-ware), none of them can be judged as being the best. Simple reason is that products are being continuously improved and no sooner than one product is adjudged the best, a more recent version of another product turns out to be slightly better.
The all in one products tend to be heavy on system resources and that's why they are generally used by non-techies. Techies prefer to install individual program from different companies that are known to be the best in that category. For ex: Zone-Alarm Firewall, AVG Anti-virus & Spybot S&D.
At the end of the day, irrespective of which protections are in force, the leak always turns out to be the human operator who let in the most recent threat into the PC with his/her unsafe practices. To be truly protected, you are the PC's only defence. Remember, the scanners can only catch what they are programmed for. You can catch with your instinct.
For starters use a safe web-browser such as Opera or Firefox. These are far less prone to web-based malware. Never open an attachment unless you know who it is from and you are expecting it. Check emails carefully for signs of infected attachments, spoofs, phishing attacks etc. When downloading from web, download via links from the original product manufacturer's website.

There is no one best product. However, there are a number of great free poducts available. Be careful searching for these, there are spyware programs that use similar names, they may delete some spyware, but install more.
Ad-Aware SE from lavasoft is probably the number one program

Two easy ones that work well in actual testing.
Windows Defender, from microsoft (www.microsoft.com)
