The Boston City sign is the famous Citgo sign.
It's the sign you see on TV all the time located in Kenmore Square, just a few minute walk from Fenway Park. It's a historical landmark located on top of one of the buildings there. And as you can see from the link below, they call it the Boston sign. :-)

Not sure what you mean by City Sign. . .If you mean a symbol of the city, it might be the skyline with the Prudential Building, or the Charles River, or even the Citgo Sign at Fenway Park. . . .The highway signs are that little Pilgrim hat outline. .. .There is no sign that says Boston though, that I can think of.

As to the Citgo sign, it used to be neon tubes. The problem is that in New England we have something called "Ice" and "Snow" which breaks glass.
It is now LED lights in plastic tubes. This makes it much more reliable, but I prefer the old tubes because the mechanical mechanism used to be more "random" that the electronic control system now used.
A few weeks ago when the Venezuelan President Hugo Chavez called Bush the Devil (In my mind pretty accurate, but I digress), a local lawmaker wanted the Citgo sign replaced with an American Flag because Citgo is partically owned by Venezuela.
