Job was a man in the Bible who had many trials and heartaches, but still held on to his faith. The story of Job consistently demonstrated how patient he was with all that had happened to him. There is a book in the Bible named after him and it tells his story.
To say someone could test the patience of Job is saying they are extremely difficult.

This long poem deals with one of the deepest problems of man: How do we explain sin and suffering, if there exists a God powerful enough to do something about them?
Theme: The mystery that surrounds human existence and the need to trust in God runs throughout the book. Mankind simply does not have enough knowledge to explain why things happen the way they do. It is possible to rise above our limitations by faith in God, however, because God does know why everything happens and will work good for those who love him. We may thus learn the profound truth that when we have nothing left but God, God is enough.

Job was a patient man who endured suffering and misery. Once he realizes that we don't need "answers" to life's problems but rather that we need God himself, Job is healed and given material and spiritual blessings far beyond what he had before.
In other words, Job was incredibly patient, and to test the patience of Job is to go beyond the ways in which God tested him.

During his lifetime, Job endured plagues, the ruin of his fields, the death of his family and much illness himself. It was a test by the devil who said he could make Job renounce God. Throughout all his suffering, Job remained faithful to God.
So, having the patience of Job means having the ability to keep going when things get really bad.

Depending on your belief, there was a man named Job and I think the Christians believe that God and Satan argued over Job's reverance to God; saying that Job only praised God because his life was so good -- so to prove a point, God plagued Job with boils, killed his crops and children. At this point, Job still praised God. So God could say to Satan - "See - told you so". So God destroyed this man's life, children and livlihood in order to prove a point to Satan.
So to test the patience of Job is a hard thing to do. He has a very high patience level.
I will NEVER understand Christians.

Read the Book of Job in the Old Testament. God loved Job very much and felt his faith was so strong, that it could be tested. It was, and Job had a whole lot of problems but he persisted. However, he had a lot of complaints to God and God responded. It is one of the most interesting stories in the Bible. I hope you read it to get the complete answer. It is Job.

I don't remember the whole story but Job - pronounced Jobe - is an Old Testiment character who wrestled with God. He was very faithful and waited years for a sign from God.
I hope there are some Biblical Scholars who can give a more in-depth answer.
