The size of the band itself or the size of the DNA pieces which are in the band?

The relative absolute size of the bands can be done by using a scanning densitometer.

The size of the DNA pieces is usually done by comparison with a standard Lambda Hind III digest or a 1KB ladder (see reference below).

By the distance they travel. Heavier bands don't travel as far as the lighter bands. They travel at knows rates, so the weight can be calculated from the distance.
