Be direct. Don't apologize, make excuses or defend your decision. Simply say that you and your roommate do not want another roommate. The more detail that you add, the more you give the person to question and perhaps put you on the defensive.
Say "No", mean it, and don't allow yourself to be drawn into a discussion about it.
